Abstract:
(EN) Using the characteristics of G-protein coupled receptors (GPCR) for sensing specific ligands and undergoing conformational change, inserting a signal molecule in an intracellular region of the G-protein coupled receptors, converting the conformational change of the G-protein coupled receptors into an optical signal change, and detecting the presence and/or concentration of a specific ligand by means of detecting the change in the optical signal; a GPCR activated fluorescent probe (GRAB probe) is constructed according to this principle. A method for using the GRAB probe to detect a specific ligand.
(FR) La présente invention concerne une sonde fluorescente activée par des récepteurs couplés à la protéine G (GPCR) (sonde GRAB), construite selon le procédé consistant : à utiliser les caractéristiques de récepteurs couplés à la protéine G destinés à détecter des ligands spécifiques et à subir un changement conformationnel, à insérer une molécule signal dans une région intracellulaire des récepteurs couplés à la protéine G, à convertir le changement conformationnel des récepteurs couplés à la protéine G en un changement de signal optique, et à détecter la présence et/ou la concentration d'un ligand spécifique par détection du changement dans le signal optique. L'invention concerne également un procédé d'utilisation de la sonde GRAB pour détecter un ligand spécifique.
(ZH) 利用G蛋白偶联受体(GPCR)感受特定配体并发生构象改变的特点,在G蛋白偶联受体的胞内区中插入信号分子,将G蛋白偶联受体的构象改变转变为光学信号的变化,通过检测光学信号的变化来检测所述特定配体的存在和/或浓度,以此为原理构建基于GPCR激活的荧光探针(GRAB探针)。以及利用GRAB探针检测特定配体的方法。
