Abstract:
(EN) A cell line capable of implementing inducible rescue of rhabdovirus, construction method therefor, and application thereof. The cell line is a BHK(TetOn) cell line for lentivirus inducible expression of T7pol/P/N. The rhabdovirus construction method comprises: transfecting dual plasmids, namely a virus polymerase gene plasmid pL and a rhabdovirus core framework plasmid pCore, into the BHK(T7pol/P/N-TetOn) cell line according to a specific ratio, thereby rapidly obtaining a recombinant rhabdovirus. A vector for inducible expression of T7pol/P/N is integrated into a BHK(TetOn) cell genome by means of pGag/Pol, pRSV-Rev, and a pMD2.G lentivirus plasmid system. The cell line can efficiently perform rapid rescue on rhabdovirus assembly and completes screening and identification, and greatly improves the rescue efficiency of a recombinant rhabdovirus as compared with the conventional multi-plasmid rescue system.
(FR) La présente invention concerne une lignée cellulaire susceptible de mettre en œuvre le sauvetage inductible de rhabdovirus, son procédé de construction, et son application. La lignée cellulaire est une lignée cellulaire BHK(TetOn) pour l’expression inductible de lentivirus de T7pol/P/N. Le procédé de construction du rhabdovirus comprend : la transfection de doubles plasmides, à savoir un plasmide de gène de polymérase viral pL et un plasmide de cadre de noyau de rhabdovirus pCore, dans la lignée cellulaire BHK(T7pol/P/N-TetOn) selon un rapport spécifique, obtenant ainsi rapidement un rhabdovirus recombinant. Un vecteur pour l’expression inductible de T7pol/P/N est intégré dans un génome cellulaire BHK(TetOn) à l’aide de pGag/Pol, pRSV-Rev, et d'un système plasmide lentiviral de pMD2.G. La lignée cellulaire peut efficacement effectuer le sauvetage rapide sur l’ensemble rhabdovirus et achève le criblage et l’identification, et améliore grandement l’efficacité du sauvetage d’un rhabdovirus recombinant tel que comparé au système de sauvetage classique multi-plasmidique.
(ZH) 一种可诱导性拯救弹状病毒的细胞系及其构建方法与应用,所述细胞系为慢病毒诱导表达T7pol/P/N的BHK(TetOn)细胞系,所述的弹状病毒构建方法是将病毒聚合酶基因质粒pL及弹状病毒核心骨架质粒pCore双质粒按照特定的比例转染BHK(T7pol/P/N-TetOn)细胞系快速获得重组弹状病毒,诱导表达T7pol/P/N的载体通过pGag/Pol,pRSV-Rev和pMD2.G慢病毒质粒系统整合于BHK(TetOn)细胞基因组。上述的细胞系能高效的对弹状病毒组装进行快速拯救并完成筛选鉴定,与传统的多质粒拯救系统相比大大的提高重组弹状病毒的拯救效率。
