Abstract:
(EN) The invention pertains to the targeted alteration of a duplex DNA in a cell, whereby two site-specific nucleases generate an indel, such that the open reading frame is not altered after the second indel. The invention further pertains to the use of such nucleases for the targeted alteration of an open reading frame in duplex DNA and a kit of parts for use in a method of the invention. Using the method of the invention, novel plants were obtained having an improved herbicide resistance. The invention therefore also concerns plants having improved herbicide resistance due to the expression of an altered ALS protein.
(FR) L'invention concerne l'altération ciblée d'un ADN duplex dans une cellule par laquelle deux nucléases spécifiques de site génèrent un indel, d'une façon telle que le cadre de lecture ouvert n'est pas modifié après le second indel. L'invention concerne en outre l'utilisation de telles nucléases pour l'altération ciblée d'un cadre de lecture ouvert dans un ADN duplex, et un kit d'éléments à utiliser dans une méthode de l'invention. L'utilisation de la méthode de l'invention a permis d'obtenir de nouvelles plantes présentant une meilleure résistance aux herbicides. L'invention concerne donc également des plantes présentant une meilleure résistance aux herbicides du fait de l'expression d'une protéine ALS modifiée.
