Abstract:
(EN) The invention belongs to the field of quality analysis and control of traditional Chinese medicine, and provides a method for separating eighteen components in a traditional Chinese medicine composition, comprising: (1) preparing the traditional Chinese medicine composition into a total extract of the traditional Chinese medicine composition, separating by resin through sequentially eluting with water, 10% ethanol and 30% ethanol, and collecting the 30% ethanol eluate to obtain a 30% ethanol extract; (2) adding the 30% ethanol extract to a reverse phase silica gel ODS-AQ-HG, and separating in a medium pressure separation column to obtain differently numbered elution dry pastes; (3) dissolving the differently numbered elution dry paste with 30% methanol as a solvent, and passing the solution through a 0.45 μm microporous membrane, carrying out a primary separation by high performance liquid chromatography and collecting chromatographic peaks with different retention times, and further purifying by high performance liquid chromatography; finally obtaining the compounds of eighteen components. The separation method of the present invention enables in-depth study of the chemical components of a pharmaceutical composition.
(FR) L'invention se rapporte au domaine de l'analyse de qualité et du contrôle de la médecine chinoise traditionnelle, et concerne un procédé de séparation de dix-huit composants dans une composition de médecine chinoise traditionnelle, comprenant les étapes consistant à : (1) préparer la composition de médecine chinoise traditionnelle en un extrait total de composition de médecine chinoise traditionnelle, séparer par résine au moyen d'une élution séquentielle avec de l'eau, 10 % d'éthanol et 30 % d'éthanol, et collecter l'éluat d'éthanol à 30 % pour obtenir un extrait d'éthanol à 30 % ; (2) ajouter l'extrait d'éthanol à 30 % à un gel de silice en phase inverse ODS-AQ-HG, et séparer dans une colonne de séparation moyenne pression pour obtenir des pâtes sèches d'élution différemment numérotées ; (3) dissoudre la pâte sèche d'élution différemment numérotée avec 30 % de méthanol en tant que solvant, et faire passer la solution à travers une membrane microporeuse de 0,45 µm, réaliser une séparation primaire par chromatographie liquide à haute performance et collecter des pics chromatographiques à différents temps de rétention, procéder à une purification supplémentaire par chromatographie liquide à haute performance ; et enfin obtenir des composés de dix-huit composants. Le procédé de séparation selon la présente invention permet d'effectuer une étude en profondeur de composants chimiques d'une composition pharmaceutique.
(ZH) 本发明属于中药的质量分析控制领域,提供一种中药组合物中十八种成分的分离方法,所述分离方法包括:(1)将中药组合物制备成中药组合物总浸膏后,经树脂分离,依次用水、10%乙醇、30%乙醇洗脱,收集30%乙醇洗脱液,得到30%乙醇浸膏;(2)将30%乙醇浸膏加入反相硅胶ODS-AQ-HG,在中压分离柱中进行分离,得到不同编号的洗脱干膏;(3)将不同编号的洗脱干膏用30%甲醇作为溶解液进行溶解,溶解液经过0.45μm微孔滤膜,采用高效液相色谱法进行初步分离,分别收集不同保留时间的色谱峰,再经高效液相色谱进一步纯化;最终制得十八种成分的化合物。本发明的分离方法能够对药物组合物的化学成分进行深入研究。
