(KO) 조현병 동물모델 및 이의 제조방법
Abstract:
(EN) The present invention relates to: a schizophrenia animal model which is a mouse in which an anoctamin 1 (ANO1) gene is specifically knocked out in cholinergic neurons of the medial habenula; and a preparation method therefor and the like. The schizophrenia animal model according to the present invention targets the medial habenula which is brain tissue playing a major role in the pathogenesis of schizophrenia, and it has been confirmed that when an ANO1 gene is specifically knocked out in the cholinergic neurons of the medial habenula, positive, negative and cognitive symptoms of schizophrenia are observed, thereby confirming that schizophrenia has been induced. Therefore, the animal model of the present invention is expected to be effectively useful in schizophrenia pathogenesis research and therapeutic agent development and screening.
(FR) La présente invention concerne : un modèle animal de schizophrénie qui est une souris dans laquelle un gène anoctamine 1 (ANO1) est spécifiquement inactivé dans les neurones cholinergiques du habenula médial ; et son procédé de préparation et similaire. Le modèle animal de schizophrénie selon la présente invention cible l'habenula médial qui est un tissu cérébral jouant un rôle majeur dans la pathogenèse de la schizophrénie et il a été confirmé que lorsqu'un gène ANO1 est spécifiquement inactivé dans les neurones cholinergiques de l'habenula médial, des symptômes positifs, négatifs et cognitifs de la schizophrénie sont observés, confirmant ainsi que la schizophrénie a été induite. Par conséquent, le modèle animal de la présente invention est censé être efficacement utile dans la recherche de la pathogenèse de la schizophrénie et le développement et le criblage d'agents thérapeutiques.
(KO) 본 발명은 내측고삐핵의 콜린성 신경세포에서 특이적으로 ANO1(Anoctamin 1) 유전자가 결손된 생쥐인 것을 특징으로 하는 조현병 동물모델 및 이의 제조방법 등에 관한 것이다. 본 발명에 따른 조현병 동물모델은 조현병 발병기전에 주요하게 작용하는 뇌조직인 내측고삐핵을 표적으로 한 것으로서, 내측고삐핵의 콜린성 신경세포에서 ANO1 유전자가 특이적으로 결손되는 경우 조현병의 양성증상, 음성증상, 및 인지증상이 나타남을 확인하여 조현병이 유발됨을 확인하였는바, 본 발명의 동물모델이 조현병의 발병기전 연구, 치료제 개발 및 스크리닝의 동물모델로 유용하게 이용될 것으로 기대된다.
