Abstract:
(EN) The present invention provides a method for purification of a sulfatase using metal chelating chromatography without use of tags (such as His-tag, etc.). An embodiment provides a method for purifying a sulfatase comprising the steps of: (a) providing a sulfatase-containing solution comprising one or a plurality of impurities; (b) performing a first chromatographic separation of the sulfatase-containing solution using a metal affinity chromatography resin; (c) performing a second chromatographic separation using a cation exchange chromatography resin; and (d) performing a final chromatographic separation using an anion exchange chromatography resin, wherein the impurities are removed thereby.
(FR) La présente invention concerne un procédé de purification d'une sulfatase à l'aide d'une chromatographie par chélation métallique sans utiliser d'étiquettes (telles que His-tag, etc.). Un mode de réalisation concerne un procédé de purification d'une sulfatase comprenant les étapes consistant à : (a) fournir une solution contenant de la sulfatase comprenant une ou plusieurs impuretés ; (b) effectuer une première séparation chromatographique de la solution contenant de la sulfatase à l'aide d'une résine de chromatographie d'affinité métallique ; (c) réaliser une seconde séparation chromatographique à l'aide d'une résine de chromatographie échangeuse de cations ; et (d) réaliser une séparation chromatographique finale à l'aide d'une résine de chromatographie par échange d'anions, les impuretés étant ainsi éliminées.
