Abstract:
(EN) The purpose of the present invention is to provide a method for producing ambrein capable of obtaining ambrein easily and efficiently. This problem can be solved by a mutant tetraprenyl-β-curcumene cyclase of the present invention in which (1) the aspartic acid which is the fourth amino acid residue of the DXDD motif is substituted by an amino acid other than aspartic acid and (2) the amino acid adjacent to the N end of the (A/S/G)RX(H/N)XXP motif is substituted by an amino acid other than tyrosine, or the fourth amino acid of the GXGX(G/A/P) motif is substituted by an amino acid other than tyrosine.
(FR) L'objet de la présente invention est de fournir une méthode de production d'ambréine capable d'obtenir facilement et efficacement de l'ambréine. Ce problème peut être résolu par une cyclase tétraprényl-β-curcuminoïde mutante de la présente invention dans laquelle (1) l'acide aspartique qui est le quatrième résidu d'acide aminé du motif DXDD est substitué par un acide aminé autre que l'acide aspartique et (2) l'acide aminé adjacent à l'extrémité N du motif (A/S/G)RX(H/N)XXP est substitué par un acide aminé autre que la tyrosine, ou le quatrième acide aminé du motif GXGX(G/A/P) est substitué par un acide aminé autre que la tyrosine.
(JA) 本発明の目的は、簡便に且つ効率的にアンブレインを得ることができるアンブレインの製造方法を提供することにある。 前記課題は、本発明の(1)DXDDモチーフの第4番目のアミノ酸残基であるアスパラギン酸がアスパラギン酸以外のアミノ酸に置換しており、(2)(A/S/G)RX(H/N)XXPモチーフのN末に隣接するアミノ酸がチロシン以外のアミノ酸に置換しているか、又はGXGX(G/A/P)モチーフの4番目のアミノ酸がロイシン以外のアミノ酸に置換している、変異型テトラプレニル-β-クルクメン環化酵素によって解決することができる。
