Abstract:
(EN) The present invention pertains to a method for detecting HBsAg with which it is possible to detect HBsAg with high sensitivity even when blood (whole blood) is used as a sample. A sample is provided on a metal film on the surface of which there is immobilized a binding substance (e.g., an antibody) capable of specifically binding to hepatitis B surface antigens, and hepatitis B surface antigens included in the sample are bound by the binding substance. The hepatitis B surface antigens are also labeled by a fluorescent substance. Fluorescence, which is emitted from the fluorescent substance when the metal film is irradiated with excitation light so that surface plasmon resonance is produced in the metal film, is detected.
(FR) La présente invention concerne un procédé de détection de l'Ag HBs au moyen duquel il est possible de détecter l'Ag HBs avec une sensibilité élevée même lorsque le sang (sang total) est utilisé comme échantillon. Un échantillon est disposé sur un film métallique sur la surface duquel est immobilisée une substance de liaison (par exemple, un anticorps) capable de se lier spécifiquement à des antigènes de surface de l'hépatite B, et des antigènes de surface de l'hépatite B présents dans l'échantillon sont liés par la substance de liaison. Les antigènes de surface de l'hépatite B sont également marqués par une substance fluorescente. La fluorescence, qui est émise à partir de la substance fluorescente lorsque le film métallique est irradié par une lumière d'excitation de telle sorte qu'une résonance plasmonique de surface est produite dans le film métallique, est détectée.
(JA) 本発明は、検体として血液(全血)を用いてもHBsAgを高感度に検出することができる、HBsAgの検出方法に関する。その表面にB型肝炎ウイルス表面抗原に特異的に結合できる結合物質(例えば抗体)が固定化された金属膜上に検体を提供して、前記検体に含まれるB型肝炎ウイルス表面抗原を前記結合物質に結合させる。また、前記B型肝炎ウイルス表面抗原を蛍光物質で標識する。前記金属膜で表面プラズモン共鳴が生じるように前記金属膜に励起光を照射したときに前記蛍光物質から放出される蛍光を検出する。
