Abstract:
(EN) Described herein is a method of method of modifying a target nucleotide sequence of cells in a population of cells, wherein said population of cells is a population of algal or plant cells, the method comprising: introducing into said cells (i) a Cpf1 endonuclease, (ii) a guide RNA and (iii) at least one oligodeoxynucleotide (ODN); and culturing the cells such that the guide RNA directs the Cpf1 endonuclease to said target nucleotide sequence and introduces a double-stranded break in said target nucleotide sequence. Also disclosed is a kit for use in the method.
(FR) L'invention concerne un procédé de modification d'une séquence nucléotidique cible de cellules dans une population de cellules, ladite population de cellules étant une population de cellules algales ou végétales, le procédé comprenant les étapes : d'introduction dans lesdites cellules (i) d'une endonucléase Cpf1, (ii) d'un ARN guide et (iii) d'au moins un oligodésoxynucléotide (ODN) ; et la culture des cellules de telle sorte que l'ARN guide dirige l'endonucléase Cpf1 vers ladite séquence nucléotidique cible et introduit une cassure bicaténaire dans ladite séquence nucléotidique cible. L'invention concerne également une trousse destinée à être utilisée dans ce procédé.
