Abstract:
(EN) The present invention generally relates to a method for analyzing and identification of the plurality of lipids in a sample that is profiled using a combined Liquid Chromatography-Mass Spectrometry (LC-MS) technique, comprising the steps of: a) providing a list of Liquid Chromatography-Mass Spectrometry (LC-MS)-based mass features; b) deconvoluting said list of LC-MS-based mass features; c) inferring daughter ions from the deconvoluted list of LC-MS-based mass features; d) identifying one or more parental exact masses from the inferred daughter ions; e) scoring each of the one more parental exact masses based on the inferred daughter ions; f) determining characteristic mass features in response to the scoring of each of the one or more parental exact masses; and g) determining each of the plurality of lipids based on the characteristic mass features thereof. In particular, the present invention also relates to identification of the plurality of lipids undergoing in-source fragmentation.
(FR) La présente invention concerne généralement un procédé d’analyse et d’identification de la pluralité de lipides dans un échantillon qui est profilé au moyen d’une technique combinée de chromatographie liquide-spectrométrie de masse (LC-MS), comprenant les étapes de : a) fourniture d’une liste de caractéristiques de masse à base de chromatographie liquide-spectrométrie de masse (LC-MS) ; b) déconvolution de ladite liste de caractéristiques de masse à base de LC-MS ; c) déduction d’ions filles à partir de la liste déconvolutée de caractéristiques de masse à base de LC-MS ; d) identification d’une ou plusieurs masses exactes parentales à partir des ions filles déduits ; e) notation de chacune des une ou plusieurs masses exactes parentales sur la base des ions filles déduits ; f) détermination de caractéristiques de masse caractéristique en réponse à la notation de chacune des une ou plusieurs masses exactes parentales ; et g) détermination de chacun de la pluralité de lipides sur la base des caractéristiques de masse caractéristique de ceux-ci. En particulier, la présente invention concerne en outre l’identification de la pluralité de lipides subissant une fragmentation dans la source.
