Abstract:
(EN) One embodiment of the present invention provides a method and an apparatus for isolating stromal cells from biological tissue without using an enzyme, the method using the moving of stromal cells to the outside of biological tissue by inducing the spontaneous migration of the stromal cells of the biological tissue, wherein the induction of the spontaneous migration of the stromal cells is carried out in a state in which the biological tissue is attached onto an attachment member made of a material to which the biological tissue can be attached, and the induction of the spontaneous migration of the stromal cells is carried out in a culture solution where the stromal cells can survive. Therefore, problems such as the toxicity and cost of an enzyme, treatment time and risks of a heterologous virus and a problem of instability caused by using an enzyme having components extracted from the gastric fluid of animals of different species can be resolved, and, simultaneously, isolation efficiency can be improved by isolating, as stromal cells of a relatively intact undamaged natural condition, stromal cells from biological tissue without using an enzyme.
(FR) Un mode de réalisation de la présente invention concerne un procédé et un appareil pour isoler des cellules stromales à partir d'un tissu biologique sans utiliser d'enzyme, le procédé utilisant le déplacement de cellules stromales vers l'extérieur d'un tissu biologique par induction de la migration spontanée des cellules stromales du tissu biologique. L'induction de la migration spontanée des cellules stromales est réalisée dans un état dans lequel le tissu biologique est fixé sur un élément de fixation constitué d'un matériau auquel le tissu biologique peut être fixé, et l'induction de la migration spontanée des cellules stromales est réalisée dans une solution de culture où les cellules stromales peuvent survivre. Par conséquent, des problèmes tels que la toxicité et le coût d'une enzyme, le temps de traitement et les risques d'un virus hétérologue, et un problème d'instabilité provoqué par l'utilisation d'une enzyme ayant des composants extraits du fluide gastrique d'animaux de différentes espèces, peuvent être résolus et, simultanément, l'efficacité d'isolement peut être améliorée en isolant, en tant que cellules stromales dans un un état naturel non endommagé et relativement intact, des cellules stromales provenant d'un tissu biologique sans utiliser d'enzyme.
(KO) 본 발명의 일 실시례는, 효소를 사용하지 않고 생체조직에서 기질세포를 분리하는 방법으로서, 생체조직의 기질세포의 자발적 마이그레이션(spontaneous migration)을 유도하여 기질세포를 생체조직의 외부로 이동시키는 것을 이용하고, 기질세포의 자발적 마이그레이션의 유도는 생체조직을 부착시킬 수 있는 소재의 부착부재 상에 생체조직이 부착된 상태에서 이루어지며, 기질세포의 자발적 마이그레이션의 유도는 기질세포가 생존할 수 있는 배양액 내에서 이루어지는 것을 특징으로 하는 방법 및 장치를 제공한다. 이에 의하여 효소의 독성과 비용, 처리 시간, 이종 바이러스에 대한 우려 등의 문제점과 이종 동물의 위액에서 추출된 성분을 갖는 효소를 사용하는 것으로 인한 불안정성의 문제점을 해소함과 동시에 생체조직으로부터 기질세포를 효소를 사용하지 않고 손상 없이 상대적으로 온전한 자연 상태의 기질세포로 분리하여 분리효율을 향상시킬 수 있다.
