Abstract:
(EN) The present disclosure provides a method for determining the undifferentiated state of a pluripotent stem cell, the method comprising irradiating a culture medium being tested in which the pluripotent stem cell is cultured, with light having wavelengths in all or a part of the range of 190-2500 nm, detecting reflected light, transmitted light, or transmitted and reflected light thereof to obtain absorbance spectral data, and analyzing the absorbances in all or in a partial range of the measured wavelengths of the absorbance spectral data, on the basis of an analysis model which is previously created using a plurality of types of control culture media used in culturing the pluripotent stem cell, to determine the undifferentiated state of the pluripotent stem cell, wherein the plurality of types of control culture media include one or more differentiation-inducing media selected from a medium used in maintaining the undifferentiated state of the pluripotent stem cell, a medium for inducing differentiation to an ectodermal cell, a medium for inducing differentiation to a mesodermal cell, and a medium for inducing differentiation to an endodermal cell.
(FR) La présente invention concerne un procédé de détermination de l'état indifférencié d'une cellule souche pluripotente, le procédé comprenant l'exposition d'un milieu de culture testé dans lequel la cellule souche pluripotente est cultivée, à une lumière ayant des longueurs d'onde dans toutes ou une partie de la plage de 190-2 500 nm, la détection de la lumière réfléchie, de la lumière transmise ou de la lumière transmise et réfléchie de celle-ci pour obtenir des données spectrales d'absorbance, et l'analyse des absorbance dans toutes les longueurs d'onde mesurées, ou une plage partielle de celles-ci, des données spectrales d'absorbance, sur la base d'un modèle d'analyse qui est préalablement créé à l'aide d'une pluralité de types de milieux de culture témoins utilisés dans la culture de la cellule souche pluripotente, pour déterminer l'état indifférencié de la cellule souche pluripotente, la pluralité de types de milieux de culture témoins comprenant un ou plusieurs milieux induisant la différenciation choisis parmi un milieu utilisé pour maintenir l'état indifférencié de la cellule souche pluripotente, un milieu pour induire la différenciation dans une cellule ectodermique, un milieu pour induire une différenciation dans une cellule mésodermique, et un milieu pour induire une différenciation dans une cellule endodermique.
(JA) 本開示は、 多能性幹細胞の未分化状態を判定する方法であって、波長190nm~2500nmの範囲またはその一部範囲の波長光を、多能性幹細胞を培養する被検培養培地に照射し、その反射光、透過光または透過反射光を検出して吸光度スペクトルデータを得、多能性幹細胞の培養に使用した複数種の対照培養培地を用いて予め作成した解析モデルに基づき、前記吸光度スペクトルデータ中の測定全波長またはその一部範囲の吸光度を解析することにより多能性幹細胞の未分化状態を判定することを含んでなり、前記複数種の対照培養培地が、多能性幹細胞の未分化状態の維持に使用した培地と、外胚葉系細胞への分化誘導培地、中胚葉系細胞への分化誘導培地および内胚葉系細胞への分化誘導培地からなる群から選択される一種以上の分化誘導培地とを含んでなる方法を提供する。
