Abstract:
(EN) Provided is application of a lettuce as a host in expressing a factor. Anti-hemophilic factor VIII (FVIII) is expressed using a recombinant vector by means of agrobacterium-mediated vacuum infiltration. The expression system determines that a plant exogenous protein can be collected after 4 d of agrobacterium infection. The successful expression of recombinant FVIII is determined by means of SDS-PAGE and Western blotting. A blood coagulation experiment proves that FVIII produced by a lettuce has biological activity. Also provided is a method for producing active recombinant anti-hemophilic FVIII.
(FR) L'invention concerne l'application d'une laitue comme hôte dans l'expression d'un facteur. Le facteur VIII anti-hémophilique (FVIII) est exprimé à l'aide d'un vecteur recombinant au moyen d'une infiltration sous vide médiée par agrobacterium. Le système d'expression détermine qu'une protéine exogène de plante peut être collectée après 4 jours d'infection par agrobacterium. L'expression réussie du FVIII recombinant est déterminée au moyen de SDS-PAGE et de transfert Western. Une expérience de coagulation sanguine prouve que le FVIII produit par une laitue a une activité biologique. L'invention concerne aussi un procédé de production de FVIII anti-hémophilique recombinant actif.
(ZH) 提供了生菜作为宿主在表达凝血因子中的应用。利用重组载体以及农杆菌介导真空渗透方法来表达人凝血八因子(anti-hemophilic factor(AHF),FVIII)。该表达体系确定植物外源蛋白在农杆菌侵染4d后就可以收集。利用SDS-PAGE法、免疫印迹法(Western法)确定重组FVIII成功表达。凝血实验证明生菜生产的FVIII具有生物学活性。还提供了一种生产具有活性重组人FVIII的方法。
