Abstract:
(EN) Disclosed are methods for performing dynamic nuclear polarization using the polarizing agents described herein. In general, the methods involve (a) providing a frozen sample in a magnetic field, wherein the frozen sample includes a polarizing agent described herein and an analyte with at least one spin half nucleus; (b) polarizing the at least one spin half nucleus of the analyte by irradiating the frozen sample with radiation having a frequency that excites electron spin transitions in the polarizing agent; (c) optionally melting the sample to produce a molten sample; and (d) detecting nuclear spin transitions in the at least one spin half nucleus of the analyte in the frozen or molten sample. In certain embodiments, the polarizing agents can be peptide-based. In these embodiments, the polarizing agents can be readily prepared by solid-phase peptide synthesis.
(FR) L'invention concerne des procédés destinés à effectuer une polarisation nucléaire dynamique à l'aide des agents de polarisation décrits ici. En général, les procédés comprennent (a) la fourniture d'un échantillon congelé dans un champ magnétique, l'échantillon congelé comportant un agent de polarisation décrit ici et un analyte doté d'au moins un noyau de spin 1/2 ; (b) la polarisation dudit noyau de spin 1/2 de l'analyte par irradiation de l'échantillon congelé avec un rayonnement présentant une fréquence qui excite des transitions de spin d'électrons dans l'agent de polarisation ; (c) la fusion éventuelle de l'échantillon en vue de produire un échantillon fondu ; et (d) la détection de transitions de spin nucléaire dans ledit noyau de spin 1/2 de l'analyte dans l'échantillon congelé ou fondu. Selon certains modes de réalisation, les agents de polarisation peuvent être à base de peptides.
