Abstract:
(EN) Methods and materials are provided for detecting nucleic acid sequence differences including single nucleotide mutations or polymorphisms, one or more nucleotide insertions, and one or more nucleotide deletions in single molecule target members present in a test population of nucleic acid fragments. Heteroduplexes are formed between members of the test nucleic acid population and their corresponding complements provided in a pool of mismatch cleavage probes. Mismatched base pairs in the heteroduplexes are specifically cleaved and cleaved probe fragments are electronically detected to signal the present of the target members in the test population.
(FR) L'invention concerne des procédés et des matériaux pour détecter des différences de séquence d'acide nucléique, comprenant des mutations ou des polymorphismes mononucléotidiques, une ou plusieurs insertions de nucléotide et une ou plusieurs délétions de nucléotides dans des éléments cibles à molécule unique présents dans une population de test de fragments d'acide nucléique. Des hétéroduplex sont formés entre des éléments de la population d'acides nucléiques de test et leurs compléments correspondants se trouvant dans un ensemble de sondes de clivage de mésappariements. Des paires de bases mésappariées dans les hétéroduplex sont spécifiquement clivées et les fragments de sonde clivés sont détectés électroniquement pour signaler la présence des éléments cibles dans la population de test.
