Abstract:
(EN) A method of assessing cytolysis of cancer cells, the method including: providing a cellsubstrate impedance monitoring cartridge having a plurality of chambers, each chamber having an electrode array configured for measuring cell-substrate impedance, wherein different chambers are preloaded with different target cells embodied as cancer cells of different lineage, origin or stage; adding effector cells to the plurality chambers for interaction with the target cells, wherein the effector cells are immune cells; monitoring cellsubstrate impedance of the plurality of chambers before and after adding the effector cells and optionally deriving an impedance-based parameter from the impedances; and determining effectiveness of effector cell killing of the different target cells by comparing the impedances or impedance-based parameters over time.
(FR) L'invention concerne un procédé d'évaluation de la cytolyse de cellules cancéreuses, le procédé comprenant : la fourniture d'une cartouche de surveillance d'impédance de substrat cellulaire possédant une pluralité de chambres, chaque chambre possédant un réseau d'électrodes conçu pour mesurer une impédance de substrat cellulaire, différentes chambres étant préchargées avec différentes cellules cibles se présentant sous la forme de cellules cancéreuses de lignée, d'origine ou de stade différents ; l'ajout de cellules effectrices à la pluralité de chambres pour une interaction avec les cellules cibles, les cellules effectrices constituant des cellules immunitaires ; la surveillance de l'impédance de substrat cellulaire de la pluralité de chambres avant et après l'ajout des cellules effectrices et la déduction facultative d'un paramètre fondé sur l'impédance à partir des impédances ; et la détermination de l'efficacité de destruction de cellules effectrices des différentes cellules cibles par la comparaison des impédances ou des paramètres fondés sur l'impédance dans le temps.
