Abstract:
(EN) The present invention relates to a method for obtaining a Bordetella pertussis-derived PRN protein, the method comprising a freezing and thawing process capable of effectively increasing an extracted level of PRN protein. In the present invention, it was found that when a pellet was frozen before treatment with urea and then slowly thawed at a cold temperature, an extracted level of PRN protein was effectively increased compared to experiment groups under other conditions. In addition, even when the method was applied to mass-scale production, a PRN extraction level was effectively increased. Therefore, a pre-treatment process for Bordetella pertussis comprising a freezing and thawing process can be effectively used for mass production of PRN for Bordetella pertussis vaccines.
(FR) La présente invention concerne un procédé d'obtention d'une protéine PRN dérivée de Bordetella pertussis, le procédé comprenant un processus de congélation et de décongélation capable d'augmenter efficacement une quantité extraite de protéine PRN. Selon la présente invention, on a découvert que lorsqu'une pastille est congelée avant le traitement avec de l'urée puis lentement décongelée à une température froide, la quantité extraite de protéine PRN est efficacement augmentée par comparaison à des groupes expérimentaux dans d'autres conditions. De plus, même lorsque le procédé est appliqué à la production en masse, la quantité extraite de PRN est efficacement augmentée. Par conséquent, un procédé de prétraitement de Bordetella pertussis comprenant un processus de congélation et de décongélation peut être utilisé efficacement pour la production en masse de PRN pour des vaccins contre Bordetella pertussis.
(KO) 본 발명은 PRN 단백질의 추출량을 효과적으로 증가시킬 수 있는 냉동 및 해동 과정을 포함하는 백일해균 유래 PRN 단백질 수득 방법에 관한 것이다. 본 발명에서 우레아 처리 전 펠렛을 냉동하고, 넁장 온도에서 서서히 해동할 경우, 다른 조건의 실험군들에 비해 PRN 단백질의 추출량이 효과적으로 증가하는 것을 확인하였다. 또한, 상기 방법을 적용한 대량생산 규모에서도 PRN 추출량이 효과적으로 증가하였다. 따라서, 냉동 및 해동 과정을 포함하는 백일해균 전처리 과정은 백일해 백신을 위한 PRN 대량 생산에 유용하게 활용될 수 있다.
