Abstract:

[Problem] To provide a method for introducing substituents into an α,β-unsaturated ketone and a prostaglandin compound synthesis method utilizing said method, the former method being for introducing substituents into the α-position and β-position of the α,β-unsaturated ketone, being utilizable for synthesizing a prostaglandin compound by a three-component linking method, and being capable of synthesizing a desired compound in high yield through a one-pot operation without requiring any of the three ingredients necessary for the synthesis to be used in large excess and without using any highly toxic heavy metal as a catalyst or using any solvent highly toxic to the living body. [Solution] The method for introducing substituents into an α,β-unsaturated ketone of the present invention is for introducing substituents onto the α-position carbon atom and β-position carbon atom of the α,β-unsaturated ketone, and is characterized by comprising a first step, in which an alkyllithium is mixed with a trialkylalkenyltin in which the tin atom has been bonded to the vinyl-position of the alkenyl group, a second step, in which the mixture obtained in the first step is mixed with a dialkylzinc, a third step, in which the mixture obtained in the second step is mixed with the α,β-unsaturated ketone, and a fourth step, in which the mixture obtained in the third step is mixed with a trifluoromethanesulfonate compound.

Le problème décrit par la présente invention est de fournir un procédé d'introduction de substituants dans une cétone α,β-insaturée et un procédé de synthèse de composé de prostaglandine utilisant ledit procédé, le procédé antérieur étant destiné à introduire des substituants dans la position α et la position β de la cétone α,β-insaturée, pouvant être utilisé pour synthétiser un composé de prostaglandine par un procédé de liaison à trois composants, et étant capable de synthétiser un composé souhaité avec un rendement élevé par l'intermédiaire d'une opération monotope sans exiger que l'un quelconque des trois ingrédients nécessaires à la synthèse ne soit utilisé en excès et sans utiliser de métal lourd hautement toxique en tant que catalyseur ou à l'aide d'un quelconque solvant hautement toxique pour le corps vivant. La solution selon la présente invention porte sur un procédé d'introduction de substituants dans une cétone α,β-insaturée destiné à introduire des substituants sur l'atome de carbone en position α et l'atome de carbone en position β de la cétone α,β-insaturée, ledit procédé est caractérisé en ce qu'il comprend une première étape, dans laquelle un alkyllithium est mélangé avec une trialkylalcényltine, l'atome d'étain étant lié à la position vinyle du groupe alcényle, une deuxième étape, dans laquelle le mélange obtenu dans la première étape est mélangé avec un dialkylzinc, une troisième étape, dans laquelle le mélange obtenu dans la deuxième étape est mélangé à la cétone α,β-insaturée, et une quatrième étape, dans laquelle le mélange obtenu dans la troisième étape est mélangé avec un composé trifluorométhanesulfonate.

【課題】α,β-不飽和ケトンのα位及びβ位へ置換基を導入方法であって、3成分連結法によるプロスタグランジン類の合成に利用可能であるのみならず、合成に必要な各3成分のいずれについて大過剰の使用量を必要とせず、また、触媒に毒性の強い重金属を用いたり、生体に対する毒性の強い溶媒を用いたりすることがなく、one-pot操作により高収率で合成するができるα,β-不飽和ケトンへの置換基導入方法、及びそれを利用したプロスタグランジン類の合成方法を提供する。 【解決手段】本発明のα,β-不飽和ケトンへの置換基導入方法は、α,β-不飽和ケトンのα位の炭素及びβ位の炭素に置換基を導入する方法であって、アルキルリチウムと、アルケニル基のビニル位にスズが結合したトリアルキルアルケニルスズとを混合する第1工程と、該第1工程の混合物とジアルキル亜鉛とを混合する第2工程と、該第2工程の混合物とα,β-不飽和ケトンとを混合する第3工程と、該第3工程の混合物とトリフルオロメタンスルホナート化合物とを混合する第4工程とを備えることを特徴とする。
