Abstract:
(EN) The present invention relates to a method for the ultra-sensitive in vitro detection and/or quantification of a substance of interest in a sample, by detecting the luminescence emission by photoluminescent inorganic nanoparticles, comprising (i) implementing photoluminescent particles comprising a photoluminescent inorganic nanoparticle formed of a crystalline matrix having at least 103 rare-earth ions, and coupled to a targeting agent for the analyte under conditions conducive to the association thereof with the analyte of the sample; (ii) excitation of the rare-earth ions of the particles by an illumination device having a power of at least 50 mW and an excitation intensity of at least 1 W/cm2; (iii) detecting the luminescence emission and (iv) determining the presence and/or concentration of the substance by interpreting said luminescence measurement. It also relates to the use of this method for in vitro diagnostic purposes , as well as an in vitro diagnostic set.
(FR) La présente invention concerne un procédé de détection et/ou de quantification ultra-sensible in vitro d'une substance d'intérêt dans un échantillon, par détection de l'émission de luminescence par des nanoparticules inorganiques photoluminescentes, comprenant (i) la mise en œuvre de particules photoluminescentes comprenant une nanoparticule inorganique photoluminescente formée d'une matrice cristalline présentant au moins 103 ions de terres rares, et couplées à un agent de ciblage de la substance à analyser, dans des conditions propices à leur association avec la substance à analyser de l'échantillon; (ii) l'excitation des ions de terres rares des particules par un dispositif d'illumination d'une puissance d'au moins 50 mW et d'une intensité d'excitation d'au moins 1 W/cm2; (iii) la détection de l'émission de luminescence et (iv) la détermination de la présence et/ou concentration de la substance par interprétation de ladite mesure de luminescence. Elle concerne également l'utilisation de ce procédé à des fins de diagnostic in vitro, ainsi qu'un ensemble de diagnostic in vitro.
