Abstract:
(EN) A method for the preparation of a three-dimensional hydrogel scaffold for cell culturing, which comprises the steps of: a) preparing at least one first uniform solid mixture comprising at least one gas-forming agent, at least one water-soluble salt and at least one non-water-soluble, water-swellable cross-linked poly(acrylic) polymer; b) preparing compressed or compact units starting from the first solid mixture; c) subjecting the compressed or compact units thus obtained to heating at a temperature in the range of 20°C-160°C, for a time of 1-120 minutes; d) leaching the thermally treated units thus obtained in an aqueous medium; e) conditioning the leached units thus obtained in water or in an aqueous solution; f) subjecting the conditioned units thus obtained to sterilization; the three-dimensional hydrogel scaffold for cell culturing obtainable with the above method comprises a non-water soluble, water-swellable cross-linked poly(acrylic) polymer, such as Carbomer or Polycarbophil, has a porosity higher than 80% and a pore size comprised between 0.1 nm and 600 μm.
(FR) L'invention concerne une méthode de préparation d'un échafaudage d'hydrogel tridimensionnel pour la culture cellulaire, qui comprend les étapes consistant à : a) préparer au moins un premier mélange solide uniforme comprenant au moins un agent producteur de gaz, au moins un sel soluble dans l'eau et au moins un polymère poly(acrylique) réticulé gonflant dans l'eau non soluble dans l'eau ; b) préparer des unités comprimées ou compactes à partir du premier mélange solide ; c) soumettre les unités comprimées ou compactes ainsi obtenues à un chauffage à une température dans la plage allant de 20°C à 160°C, pendant une durée de 1 à 120 minutes ; d) lixivier les unités traitées thermiquement ainsi obtenues dans un milieu aqueux ; e) conditionner les unités lixiviées ainsi obtenues dans de l'eau ou dans une solution aqueuse ; f) soumettre les unités conditionnées ainsi obtenues à une stérilisation ; l'échafaudage d'hydrogel tridimensionnel pour la culture cellulaire pouvant être obtenu avec la méthode ci-dessus comprend un polymère poly(acrylique) réticulé gonflant dans l'eau, non-soluble dans l'eau, tel qu'un Carbomère ou un Polycarbophile, a une porosité supérieure à 80 % et une taille de pore comprise entre 0,1 nm et 600 µm.
