Abstract:
(EN) An aspartase variant, a preparation method therefor, and an application thereof are provided. The amino acid sequence of the aspartase variant has 96% or higher identity with sequence 2 in the sequence listing, comprises T187I and N326C mutations at positions 187 and 326 of sequence 2, respectively, and has improved catalytic acrylic acid amino-adding activity compared with the aspartase shown in sequence 2. Experiments prove that the aspartase variant can improve catalytic acrylic acid amino-adding activity compared with a wild-type parent aspartase, and has better thermal stability and pH spectrum, and can greatly improve the conversion of acrylic acid in acrylic acid amino-adding reactions.
(FR) L'invention concerne une variante d'aspartase, un procédé de préparation de celle-ci, et une application de celle-ci. La séquence d'acides aminés de la variante d'aspartase présente 96 % ou plus d'identité avec la séquence 2 dans la liste de séquences, comprend des mutations T187I et N326C aux positions 187 et 326 de la séquence 2, respectivement, et présente une activité d'ajout d'acide acrylique catalytique améliorée par rapport à l'aspartase représentée en séquence 2. Des expériences prouvent que la variante d'aspartase peut améliorer l'activité d'ajout d'acide acrylique catalytique par comparaison avec une aspartase parente de type sauvage, et présente une meilleure stabilité thermique et un meilleur spectre de pH, et peut grandement améliorer la conversion de l'acide acrylique dans des réactions d'ajout d'acide acrylique.
(ZH) 提供了一种天冬氨酸酶变体及其制备方法与应用。所述天冬氨酸酶变体其氨基酸序列与序列表中序列2相比具有96%以上同一性,且在序列2的第187位和326位分别存在T187I和N326C的突变,并且与序列2所示的天冬氨酸酶相比具有改善的催化丙烯酸加氨活性。实验证明,所述天冬氨酸酶变体与野生型亲本天冬氨酸酶相比确实具有改善的催化丙烯酸加氨活性,且热稳定性和pH谱更优,在丙烯酸加氨反应中可以大大提高丙烯酸的转化率。
