Abstract:
(EN) Disclosed is a method for capturing target cells or molecules in solution, comprising steps of: (I) getting medium containing said target cells or molecules into an apparatus comprising a capturing device for capturing said target cells or molecules; (II) getting said medium flow through said capturing device; (III) removing unbound debris, cells and molecules; (IV) getting said target cells or molecules detached from the capturing device; and (V) collecting said target cells or molecules; wherein said capturing device comprises at least one functionalized mesh comprising a mesh substrate and a functional layer formed on said mesh substrate, wherein said functional layer comprises capturing substances that can specifically bind with said target cells or molecules. The method has high specificity, as well as high throughput, and is suitable for capturing cells or molecules in a solution or expressed at the surface of cell membranes. It is particularly suited to capture and sort circulating tumor cells.
(FR) L'invention concerne un procédé de capture de cellules ou de molécules cibles en solution, comprenant les étapes consistant à : (I) introduire un milieu contenant lesdites cellules ou molécules cibles dans un appareil comprenant un dispositif de capture pour capturer lesdites cellules ou molécules cibles; (II) amener ledit milieu à s'écouler à travers ledit dispositif de capture; (III) éliminer les débris, les cellules et les molécules non liés; (IV) amener lesdites cellules ou molécules cibles à se détacher du dispositif de capture; et (V) collecter lesdites cellules ou molécules cibles; ledit dispositif de capture comprenant au moins un maillage fonctionnalisé comprenant un substrat maillé et une couche fonctionnelle formée sur ledit substrat maillé, ladite couche fonctionnelle comprenant des substances de capture qui peuvent se lier de manière spécifique auxdites cellules ou molécules cibles. Le procédé présente une spécificité élevée, ainsi qu'un rendement élevé, et est approprié pour capturer des cellules ou des molécules dans une solution ou exprimées à la surface de membranes cellulaires. Il est particulièrement approprié pour capturer et trier des cellules tumorales circulantes.
