Abstract:
(EN) The invention refers to a recombinant Pseudomonas putida host cell capable of producing D-xylonate (D-xylonic acid) from D-xylose and a method for producing D-xylonate using the same. The recombinant P. putida host cell according to the present invention is, preferably, a P. putida EM42 strain which has been genetically modified to comprise and express a heterologous beta-glucosidase gene encoding a β-glucosidase enzyme from, preferably, the thermophilic Gram-positive bacterium Thermobifida fusca. This host cell of the invention is capable of growing in a minimal mineral culture medium comprising cellobiose as a sole carbon source. When applying the recombinant P. putida of the invention, it is possible to produce D-xylonate from D-xylose with high yield while using D-cellobiose as a sole carbon source for bacterial growth.
(FR) L'invention concerne une cellule hôte de Pseudomonas putida recombinante capable de produire du D-xylonate (acide D-xylonique) à partir de D-xylose, et un procédé de production de D-xylonate utilisant ladite cellule hôte. La cellule hôte P. putida recombinante selon la présente invention est, de préférence, une souche de P. putida EM42 qui a été génétiquement modifiée de manière à comprendre et à exprimer un gène de β-glucosidase hétérologue codant pour une enzyme de β-glucosidase, de préférence, de la bactérie à Gram positif thermophile Thermobifida fusca. Cette cellule hôte selon l'invention est capable de croître dans un milieu de culture minéral minimal comprenant du cellobiose comme unique source de carbone. Lors de l'application de la P. putida recombinante de l'invention, il est possible de produire du D-xylonate à partir de D-xylose à un rendement élevé tout en utilisant du D-cellobiose comme unique source de carbone pour la croissance bactérienne.
