Abstract:
(EN) The present invention is directed to methods, compositions and devices useful for the detection and/or quantification of a microbial contaminant, including an endotoxin. In embodiments, cartridges are provided that include dried compositions that are useful in absorbance-based assays and in combination with portable readers/ devices. The cartridges involve a pump mechanism, e.g. a two position syringe, wherein a first position creates a vacuum to introduce the sample into the fluid inlet port and the conduit of the cartridge, and a second position provides transport from the conduit to an optical sample well, which contains a dried hemocyte lysate composition and a dried chromogenic substrate.
(FR) La présente invention concerne des procédés, des compositions et des dispositifs utiles pour la détection et/ou la quantification d'un contaminant microbien, comprenant une endotoxine. Dans des modes de réalisation, l'invention concerne des cartouches qui comprennent des compositions séchées qui sont utiles dans des dosages basés sur l'absorbance et en combinaison avec des lecteurs/dispositifs portables. Les cartouches comprennent un mécanisme de pompe, par exemple une seringue à deux positions, une première position créant un vide pour introduire l'échantillon dans l'orifice d'entrée de fluide et le conduit de la cartouche, et une seconde position produisant un transport du conduit à un puits d'échantillon optique, qui contient une composition de lysat d'hémocyte séchée et un substrat chromogène séché.
