Abstract:
(EN) Aspects of the disclosure relate to methods and synthetic regulatory systems for more efficient nuclease-mediated homology-directed repair (HDR). In particular, provided herein are methods for more efficient in vivo and in vitro HDR-based gene editing where the methods comprise introducing into a cell a synthetic regulatory system comprising Cas nuclease, guide RNAs (gRNAs) having various lengths and configured to target distinct nucleotide sequences for simultaneous transcriptional repression (or activation) and genome editing via double stranded break and use of a donor nucleic acid molecule as a template for repair.
(FR) Des aspects de l'invention concernent des procédés et des systèmes de régulation de synthèses pour une réparation dirigée par une homologie (HDR) plus efficace, médiée par une nucléase. En particulier, on propose ici des procédés d'édition génétique à base de HDR, in vivo et in vitro, plus efficaces, les procédés comprenant l'introduction dans une cellule d'un système de régulation synthétique comprenant la nucléase Cas, des ARN guides (gARN) ayant diverses longueurs et étant conçus pour cibler des séquences cibles distinctes de nucléotides pour une répression (ou pour une activation) simultanée de transcription et pour une édition de génome par rupture des deux brins et par usage d'une molécule d'acide nucléique donneur en tant que matrice de réparation.
