Abstract:
(EN) Disclosed are methods for the identification and production of highly efficient ("small interfering RNAs") in cytoplasmic extracts of Nicotiana tabacum. The system allows the reconstitution of antiviral RNA-silencing in vivo. More particularly, all DCL (Dicer-like proteins) and AGO (Argonaute) proteins are active in the extracts described. Such siRNAs thus provided are used in gene regulation in target organisms, inter alia in the improvement of pathogen resistance.
(FR) L'invention concerne des méthodes d'identification et de production de siARN à efficacité élevée ("petits ARN interférents" ) dans des extraits cytoplasmiques de Nicotiana tabacum. Ce système permet la reconstitution du silençage d'ARN antiviral in vivo. En particulier, toutes les protéines DCL (Dicer) et AGO (Argonautes) sont actives dans les extraits décrits. Les siARN ainsi obtenus trouvent des applications dans la régulation génique dans des organismes cibles, entre autres pour l'amélioration de la résistance aux pathogènes.
(DE) Es werden Methoden für die Identifikation und Produktion von hoch effizienten siRNAs („small interfering RNAs") in zytoplasmatischen Extrakten von Nicotiana tabacum bereitgestellt. Das System erlaubt die Rekonstitution des antivitalen RNA-silencings in vivo. Im Besonderen sind in den beschriebenen Extrakten alle DCL (Dicer) und AGO (Argonaute) Proteine aktiv. Solchermassen bereitgestellte siRNAs finden Anwendung in der Genregulation in Zielorganismen, unter anderem in der Verbesserung von Pathogenresistenz.
