Abstract:
(EN) Disclosed in the present application are a method for producing an enzymatic reaction by using adenosine to replace adenosine triphosphate (ATP) and an application thereof. The method comprises the following steps: (1) in an enzymatic reaction system, adding an ATP regenerating enzyme, adenylate kinase (AK) enzyme and adenosine in proportion to carry out an enzymatic reaction; (2) directly separating an immobilized ATP regenerating enzyme and AK enzyme in a reaction tank, and separating a free ATP regenerating enzyme and AK enzyme by means of an ultrafiltration membrane in a filter; (3) separating and purifying the filtrate obtained in step (2) to obtain a product. The present invention has the following beneficial effects: 1) Using adenosine to replace ATP or adenosine monophosphate (AMP) saves a large amount of costs in industrial production; 2) a stable enzyme recovery system is established, thus saving energy and being environmentally friendly; 3) a small amount of generated by-product ATP, adenosine diphosphate (ADP) and AMP may be directly used in a cyclic reaction, or used to produce ATP, or concentrated and purified by means of filtration, ion exchange and like methods, while operation is simple.
(FR) La présente invention concerne un procédé de production d'une réaction enzymatique à l'aide d'adénosine pour remplacer l'adénosine triphosphate (ATP) et une application de celle-ci. Le procédé comprend les étapes suivantes : (1) dans un système de réaction enzymatique, l'ajout d'une enzyme de régénération d'ATP, d'une enzyme d'adénylate kinase (AK) et d'adénosine en proportion pour effectuer une réaction enzymatique ; (2) la séparation directe d’une enzyme de régénération d'ATP immobilisée et d’une enzyme AK dans un réservoir de réaction, et la séparation d’une enzyme de régénération ATP libre et d’une enzyme AK au moyen d'une membrane d'ultrafiltration dans un filtre ; (3) la séparation et la purification du filtrat obtenu à l'étape (2) pour obtenir un produit. La présente invention présente les effets bénéfiques suivants : 1) l'utilisation d'adénosine pour remplacer l'ATP ou l'adénosine monophosphate (AMP) permet de réaliser des économies importantes dans la production industrielle ; 2) un système de récupération d'enzyme stable est établi, économisant ainsi de l'énergie et étant respectueux de l'environnement ;3) une petite quantité de sous-produit généré par l’ATP, de l'adénosine diphosphate (ADP) et de l'AMP peut être directement utilisée dans une réaction cyclique, ou utilisée pour produire de l'ATP, ou concentrée et purifiée au moyen d'une filtration, d'un échange d'ions et de procédés similaires, tout en ayant un fonctionnement simple.
(ZH) 本发明公开了一种利用腺苷代替ATP进行酶促反应生产的方法及其应用,该方法包括以下步骤:(1)在酶促反应体系中,按比例添加ATP再生酶、AK酶以及腺苷进行酶促反应;(2)固定化的ATP再生酶和AK酶在反应罐中直接分离,游离的ATP再生酶和AK酶通过过滤器中超滤膜分离;(3)将步骤(2)滤出液进行分离、纯化,获得产物。本发明具有如下有益效果:1)使用腺苷替代ATP或AMP,为工业生产节约了大量成本;2)建立了稳定的酶回收体系,节能环保;3)少量生成的副产物ATP、ADP及AMP或直接用于循环反应,或用于生产ATP,或集中通过过滤、离子交换等方法进行纯化,操作简单。
