Abstract:
(EN) The present disclosure provides compositions, methods, and systems for targeted plant genome editing. In some embodiments, the present disclosure provides recombinant self-replicating RNAs derived from a plant virus vector, such as those derived from Tobacco Mosaic Virus (TMV). In some embodiments, the recombinant self-replicating RNAs direct the expression of both a CRISPR endonuclease and a guide RNA capable of directing sequence-specific binding of the CRISPR endonuclease to a target DNA in the genome of plant cell.
(FR) La présente invention concerne des compositions, des procédés et des systèmes pour l'édition génomique de plante ciblée. Dans certains modes de réalisation, la présente invention concerne des ARN autoréplicatifs recombinés dérivés d'un vecteur viral végétal, tels que ceux dérivés du virus de la mosaïque du tabac (VMT). Dans certains modes de réalisation, les ARN autoréplicatifs recombinés dirigent l'expression d'une endonucléase CRISPR et d'un ARN guide capable de diriger la liaison spécifique d'une séquence de l'endonucléase CRISPR à un ADN cible dans le génome d'une cellule végétale.
