Abstract:
The purpose of the present invention is to provide a modified Cas9 protein having relaxed restriction of the target sequence while maintaining the ability to bind with guide RNA and the use thereof. A protein or the like including an amino acid sequence in which the arginine 1335 in SEQ ID NO: 1 is mutated to alanine (R1335A), isoleucine (R1335I), methionine (R1335M), threonine (R1335T), or valine (R1335V), the leucine 1111 is mutated to arginine (L1111R), the aspartic acid 1135 is mutated to valine (D1135V), the glycine 1218 is mutated to arginine (G1218R), the glutamic acid 1219 is mutated to phenylalanine (E1219F), the alanine 1322 is mutated to arginine (A1322R), and the threonine 1337 is mutated to arginine (T1337R).
