Abstract:
Provided is a lymphoblastic leukaemia CAR-T therapeutic vector based on OCTS technology, comprising a lentiviral skeleton plasmid, a human EF1α promoter, an OCTS chimeric receptor structural domain and IL6R single chain antibodies, the OCTS chimeric receptor structural domain comprising: a CD8 leader chimeric signal peptide; two groups of single chain antibodies, the first group being any one group selected from the following four groups of single chain antibodies: CD20 single chain antibody light chain VL and CD20 single chain antibody heavy chain VH; CD22 single chain antibody light chain VL and CD22 single chain antibody heavy chain VH; CD30 single chain antibody light chain VL and CD30 single chain antibody heavy chain VH; and CD123 single chain antibody light chain VL and CD123 single chain antibody heavy chain VH, and the second group being CD19 single chain antibody light chain VL and CD19 single chain antibody heavy chain VH; an antibody Inner-Linker; a single chain antibody Inter-Linker; a CD8 Hinge chimeric receptor hinge; a CD8 Transmembrane chimeric receptor transmembrane region; a TCR chimeric receptor T-cell activation domain; and a chimeric receptor costimulatory factor region. Also provided are a construction method for the vector and an application thereof for preparing a drug for the treatment of lymphoblastic leukaemia.
