Title: METHOD FOR STEPWISE ASSEMBLING DNA ORIGAMI UNITS
Abstract:
A method for stepwise assembling nucleic acid origami units into a nucleic acid nanostructure, comprising the following steps: 1) preparing a first-step target nucleic acid origami unit group to a m-th-step target nucleic acid origami unit group used in the following step 2) or step 3) according to a pre-formed nucleic acid nanostructure, m being an integer greater than or equal to 2; 2) stepwise linking and fixing each of the target nucleic acid origami unit groups obtained in step 1) to a stationary phase linked with a linker chain, so as to obtain a nucleic acid nanostructure linked with the linker chain; and 3) adding a separation chain which is completely complementary to the linker chain to the nucleic acid nanostructure linked with the linker chain obtained in the step 2) for separation reaction, so as to obtain said nucleic acid nanostructure. The present method achieves successful assembly of more structural monomers, demonstrating the high self-assembly efficiency of this novel method.
