Abstract:
(EN) Provided are chemical inducers of lineage reprogramming (CiLR) which include glycogen synthase kinase inhibitors, TGFβ receptor inhibitors, cyclic AMP agonists or histone acetylators. Also provided is a method of inducing lineage reprograming in a partially or completely differentiated cell of a first type into a cell with characteristics of a second and different lineage. The method includes: contacting a cell with the CiLR for a sufficient period of time to result in reprograming the cell into a modified XEN-like cell which is subsequently programmed into a cell with characteristics of a second and different lineage.
(FR) L'invention concerne des inducteurs chimiques de la reprogrammation de lignées (CiLR) qui comprennent des inhibiteurs de la glycogène synthase kinase, des inhibiteurs des récepteurs du TGFβ, des agonistes de l'AMP cyclique ou des acétylateurs d'histone. L'invention concerne également un procédé d'induction d'une reprogrammation de lignée dans une cellule partiellement ou complètement différenciée d'un premier type dans une cellule avec des caractéristiques d'une seconde lignée, différente. Le procédé comprend les étapes suivantes : la mise en contact d'une cellule avec les CiLR pendant une durée suffisante pour conduire à la reprogrammation de la cellule en une cellule de type XEN modifiée qui est ensuite programmée en une cellule avec des caractéristiques d'une seconde lignée, différente.
