Abstract:
Disclosed are methods for mitigating vaccine-enhanced infection, such as vaccinations against a flavivirus such as Zika which can lead to enhanced infection of other viruses such as Chikungunya virus. Also disclosed are chimeric adenoviruses deleted for the E1A/B region wherein flavivirus proteins are included for vaccination. Also disclosed are chimeric adenoviruses comprising a Zika sequence, such as a full length prM-E ectodomain fusion region or an E gene deleted for a conserved fusion-loop epitope. Deletion of this epitope mitigates cross-infectivity.
