Abstract:
(EN) The present invention relates to diacylglycerol acyltransferase genes and proteins, and methods of their use. In particular, the invention describes genes encoding proteins having increased diacylglycerol acetyltransferase activity compared to prior art proteins, specifically for transferring an acetyl group to a diacylglycerol substrate to form acetyl-Triacylglycerols (acetyl-TAGS), for example, a 3-acetyl-1,2-diacyl-sn-glycerol. The present invention encompasses both native and recombinant wild-type forms of the transferase, as well as mutants and variant forms. The present invention also relates to methods of using the diacylglycerol acyltransferase genes and proteins, including their expression in transgenic organisms at commercially viable levels, for increasing production of 3-acetyl-1,2-diacyl-sn-glycerols in plant oils and altering the composition of oils produced by microorganisms, such as yeast, by increasing acetyl-TAG production. Additionally, oils produced by methods of the present inventions comprising genes and proteins are contemplated for use as biodiesel fuel, in polymer production and as naturally produced food oils with reduced calories.
(FR) La présente invention concerne des gènes et des protéines de diacylglycérol acyltransférase et leurs procédés d'utilisation. En particulier, l'invention porte sur des gènes codant pour des protéines ayant une activité diacylglycérol acétyltransférase accrue par rapport aux protéines de l'état de la technique, en particulier pour le transfert d'un groupe acétyle vers un substrat diacylglycérol afin de former des acétyl-triacylglycérols (acétyl-TAG), par exemple un 3-acétyl-1,2-diacyl-sn-glycérol. La présente invention comprend les formes de type sauvage à la fois natives et recombinantes de la transférase, ainsi que les formes mutantes et variantes. La présente invention porte également sur des procédés d'utilisation des gènes et des protéines de diacylglycérol acyltransférase, comprenant leur expression dans des organismes transgéniques à des taux commercialement viables, pour augmenter la production de 3-acétyl-1,2-diacyl-sn-glycérols dans des huiles végétales et modifier la composition d'huiles produites par des microorganismes, tels qu'une levure, par augmentation de la production d'acétyl-TAG. De plus, les huiles obtenues par les procédés de la présente invention comprenant des gènes et protéines sont envisagées pour être utilisées comme carburant biodiesel, dans la production de polymères et comme huiles alimentaires produites de façon naturelle à faible teneur en calories.
