Abstract:
(EN) [Problem] To provide a cellular preparation containing highly therapeutically effective mesenchymal stem cells (MSCs). [Solution] The present invention provides a method for producing activated MSCs, the method comprising a step for using a cell culture carrier having a three-dimensional structure consisting of fibers to culture MSCs in a medium containing an activating agent derived from a mammalian fetal appendage. Also, provided are: a therapeutic effect marker of MSCs, wherein the marker is selected from the group consisting of p16ink4a, p14ARF, CDK4, CDK6, RB, and CD47; a method for determining therapeutic effects using said marker; a method for determining the adaptability of MSCs with respect to a treatment for enhancing therapeutic effects; a cell preparation containing MSCs; and a method for producing the cell preparation.
(FR) Le problème abordé par la présente invention est de pourvoir à une préparation cellulaire contenant des cellules souches mésenchymateuses (MSC) très efficaces sur le plan thérapeutique. La solution selon l'invention porte sur un procédé de production de MSC activées, où le procédé comprend une étape d'utilisation d'un support de culture cellulaire ayant une structure tridimensionnelle constituée de fibres pour cultiver les MSC dans un milieu contenant un agent d'activation dérivé d'un appendice fœtal de mammifère. Un marqueur d'effets thérapeutiques des MSC, où le marqueur est choisi dans le groupe constitué par p16ink4a, p14ARF, CDK4, CDK6, RB, et CD47; une méthode de détermination des effets thérapeutiques à l'aide dudit marqueur; une méthode de détermination de l'adaptabilité des MSC par rapport à un traitement destiné à améliorer lesdits effets thérapeutiques; une préparation cellulaire contenant des MSC; et un procédé de production de la préparation cellulaire sont en outre décrits.
(JA) 【課題】 本発明は、治療効果の高いMSCを含む細胞製剤を提供することを目的とする。 【解決手段】 ファイバーからなる3次元構造を有する細胞培養担体を用いて、哺乳動物胎児付属物由来の賦活化剤を含む培地中でMSCを培養する工程を含む、賦活化されたMSCの製造方法が提供される。また、p16ink4a、p14ARF、CDK4、CDK6、RB及びCD47よりなる群から選択されるMSCの治療効果マーカー、同マーカーを用いた治療効果判定方法、治療効果を高めるための処理に対するMSCの適応性判定方法、MSCを含む細胞製剤及びその製造方法も提供される。
