Abstract:
(EN) Provided are a method and a device for forming clusters of amplified nucleic acid fragments in a regular arrangement on a substrate without amplification bias. The method according to the present invention comprises: forming liquid droplets, in which a nucleic acid serving as a template is enclosed, on a substrate, said substrate having a plurality of first surfaces having a hydrophilic nature and second surfaces respectively surrounding the first surfaces and having a less hydrophilic nature than the first surfaces; performing a nucleic acid amplification reaction in the liquid droplets on the substrate; removing the liquid droplets; and then further performing the nucleic acid amplification reaction on the substrate.
(FR) L'invention concerne un procédé et un dispositif de formation de groupes de fragments d'acide nucléique amplifiés selon un agencement régulier sur un substrat sans biais d'amplification. Le procédé selon la présente invention consiste à : former des gouttelettes de liquide, dans lesquelles un acide nucléique servant de modèle est enfermé, sur un substrat, ledit substrat ayant une pluralité de premières surfaces ayant une nature hydrophile et secondes surfaces entourant respectivement les premières surfaces et ayant une nature moins hydrophile que les premières surfaces ; réaliser une réaction d'amplification de l'acide nucléique dans les gouttelettes de liquide sur le substrat ; éliminer les gouttelettes de liquide ; et ensuite réaliser de nouveau la réaction d'amplification de l'acide nucléique sur le substrat.
(JA) 基板上に規則的な配置で、増幅核酸断片のクラスタを増幅バイアスなく形成するための方法および装置を提供する。本発明に係る方法では、親水性を有する複数の第一の表面と、前記複数の第一の表面のそれぞれを囲む、前記第一の表面より親水性が低い第二の表面とを有する基板上で、鋳型となる核酸が封入された液滴を形成し、基板上の液滴内で核酸増幅反応を行った後、液滴を取り除いてさらに基板上で核酸増幅反応を行う。
