Title: METHOD AND SYSTEM FOR COMBINED ASSEMBLY OF SECOND GENERATION SEQUENCE AND THIRD GENERATION SEQUENCE GENOMES
Abstract:
Disclosed are a method and system for combined assembly of second generation sequence and third generation sequence genomes. The method comprises the following steps: (1) extracting a sample genome and breaking with ultrasound; (2) gel purifying fragments broken with ultrasound in step (1) and cutting the gel so as to construct libraries of different insert fragments; (3) performing second generation paired-end sequencing on the libraries produced in step (2); (4) splicing read 1 and read 2 of the second generation paired-end sequencing of the libraries; (5), performing sequence assembly on the spliced sequences; and (6) comparing a contiguous group produced in step (5) to third generation sequence data and performing combined assembly, where the sequences of read 1 and read 2 of the second generation paired-end sequencing have a contiguous area of 5 bp or more. The present method and system extend the average length of second generation sequences.
