The invention relates to an in vitro method of rescuing negative RNA virus from low virulent virus strain(s) or lentogenic-like virus strain(s) belonging to the order Mononegavirales including Bomaviridae, Filoviridae, Mymonaviridae, Nyamiviridae, Paramyxoviridae, Pneumoviridae, and Rhabdoviridae families, preferably belonging to the Paramyxoviridae family, comprising at least the steps of: (i) co-transfecting host cells with a two-plasmid system comprising a. a pGenome plasmid comprising at least a sequence encoding a virus genome from low virulent virus strain(s) or lentogenic-like virus strain(s) and b. a pNPL helper plasmid comprising at least the sequences encoding the structural viral proteins nucleocapsid protein (N), phosphoprotein (P) and large protein (L), (ii) culturing host cells under conditions for replication and transcription of the virus, and (iii) recovering the rescued negative strand RNA viruses.

L'invention concerne un procédé in vitro de sauvetage d'un virus à ARN négatif d'une ou plusieurs souches virales de faible virulence ou d'une ou plusieurs souches virales de type lentogène appartenant à l'ordre Mononegavirales, comprenant les familles Bomaviridae, Filoviridae, Mymonaviridae, Nyamiviridae, Paramyxoviridae, Pneumoviridae, et Rhabdoviridae, de préférence appartenant à la famille Paramyxoviridae, comprenant au moins les étapes suivantes : (i) co-transfection de cellules hôtes avec un système à deux plasmides comprenant a. un plasmide pGenome comprenant au moins une séquence codant pour un génome viral d'une ou plusieurs souches virales de faible virulence ou d'une ou plusieurs souches virales de type lentogène et b. un plasmide auxiliaire pNPL comprenant au moins les séquences codant pour les protéines virales structurales protéine nucléocapsidique (N), phosphoprotéine (P) et grande protéine (L), (ii) culture des cellules hôtes dans des conditions de réplication et de transcription du virus, et (iii) récupération des virus à ARN à brin négatif sauvés.
