Abstract:
(EN) Detection of under-expression of selected genes in NK cells is used to confirm a hypoxic tumor microenvironment that is ordinarily suppressive with respect to ADCC and cytotoxic cell killing of NK cells. Most notably, while hypoxia is known to upregulate HIF-1α and genes under the control of HIF-1α, hypoxia in a tumor microenvironment led to under-expression of selected genes, including HIF-1α. Thus, gene expression analysis of certain genes on NK cells can be used to detect conditions in a tumor that would indicate use of haNK cells.
(FR) La présente invention concerne la détection d'une sous-expression de gènes sélectionnés dans des cellules NK qui est utilisée pour confirmer un micro-environnement tumoral hypoxique qui est normalement suppresseur par rapport à l'ADCC et à la destruction de cellules cytotoxiques de cellules NK. Plus particulièrement, tandis que l'hypoxie est connue pour réguler à la hausse l'HIF -1α et des gènes sous le contrôle de l'HIF -1α, l'hypoxie dans un microenvironnement tumoral conduit à une sous-expression de gènes sélectionnés, y compris l'HIF -1α. Ainsi, une analyse d'expression génique de certains gènes sur des cellules NK peut être utilisée pour détecter des états d'une tumeur qui indiqueraient l'utilisation de cellules haNK.
