Abstract:
(EN) Provided herein are methods and compositions for reducing probe count variability of a biological sample. After obtaining a biological sample, such as a saliva sample, the sample is pretreated with a lysis buffer that includes a detergent to form an extraction solution. Nucleic acids are isolated from the extraction solution and fragmented into polynucleotide fragments, which are then mixed with homologous capture probes, which, for example, are bound to a flow cell of a direct targeted sequencing system. The capture probes bind to targeted sequences of the polynucleotide fragments, thereby capturing the targeted polynucleotide fragments. Based on binding of polynucleotides fragments to the homologous capture probes, a probe count is determined for the homologous probes. By mixing the lysis buffer with the biological sample to form the extraction solution, variability of the determined probe count is substantially reduced.
(FR) L'invention concerne des procédés et des compositions pour réduire la variabilité du nombre de sondes d'un échantillon biologique. Après obtention d'un échantillon biologique, tel qu'un échantillon de salive, l'échantillon est prétraité avec un tampon de lyse qui comprend un détergent pour former une solution d'extraction. Les acides nucléiques sont isolés de la solution d'extraction et fragmentés en fragments polynucléotidiques, qui sont ensuite mélangés à des sondes de capture homologues, qui, par exemple, sont liées à la cellule à flux d'un système de séquençage à ciblage direct. Les sondes de capture se lient à des séquences ciblées des fragments polynucléotidiques, pour capturer ainsi les fragments polynucléotidiques ciblés. En fonction de la liaison des fragments polynucléotidiques aux sondes de capture homologues, un nombre de sondes est déterminé en termes de sondes homologues. Le mélange du tampon de lyse avec l'échantillon biologique pour former la solution d'extraction permet de sensiblement réduire la variabilité du nombre de sondes déterminé.
