Abstract:
(EN) Provided is a method for transdifferentiating non-neuronal cells into neuronal cells, comprising subjecting an extracellular matrix-skeleton system of the non-neuronal cells to an interference treatment. The interference treatment is selected from: treatment with small-molecule inhibitors of cytoskeletal proteins, a knockdown treatment of the expression of specific genes of the extracellular matrix-skeleton system by using small interfering RNA (siRNA), and a low-adhesion treatment and the directed differentiation culture of the extracellular matrix. This transdifferentiation method can be used for tissue regeneration, repair and tumour therapy.
(FR) L'invention concerne un procédé de transdifférenciation de cellules non neuronales en cellules neuronales, comprenant la soumission d'un système de squelette matriciel extracellulaire des cellules non neuronales à un traitement d'interférence. Le traitement d'interférence est choisi parmi : un traitement avec des inhibiteurs à petites molécules de protéines cytosquelettiques, un traitement d'inactivation de l'expression de gènes spécifiques du système de squelette matriciel extracellulaire à l'aide de petits ARN interférents (ARNsi) et un traitement à faible adhérence et la culture de différenciation dirigée de la matrice extracellulaire. Ce procédé de transdifférenciation peut être utilisé pour la régénération et la réparation tissulaires et la thérapie tumorale.
(ZH) 提供了一种将非神经元细胞转分化为神经元细胞的方法,包括对非神经元细胞的胞外基质-骨架系统进行干扰处理,该干扰处理选自:采用细胞骨架蛋白小分子抑制剂进行处理,采用小干扰RNA(siRNA)对胞外基质-骨架系统的特定基因表达进行敲低处理,对胞外基质进行低粘附处理并定向分化培养。该转分化的方法可用于组织再生、修复和肿瘤治疗。
