Abstract
(EN) Provided herein are engineered systems and methods for establishing DNA adenine methylation at specific genomic locations and using DNA adenine methylation as an artificial chemical "handle" on the genome. These systems and methods allow for placing the handle on specific genomic locations as well as molecular technologies to bind, spatially spread, and maintain the handle. The systems described herein comprise, in some embodiments, three functional modules that mediate m6A operations: (1) a synthetic initiator module to place m6A at specific genomic sites; (2) a synthetic readout module to program m6A recognition and m6A-dependent transcriptional logic; and (3) propagation module that implements read-write, a mechanism proposed to underlie chromatin spreading and epigenetic maintenance across cellular systems.
(FR) L'invention concerne des systèmes et des procédés modifiés permettant d'établir une méthylation d'adénine d'ADN à des emplacements génomiques spécifiques et d'utiliser la méthylation de l'adénine d'ADN comme "manœuvre" chimique artificielle sur le génome. Ces systèmes et procédés permettent de placer la manipulation sur des emplacements génomiques spécifiques ainsi que des technologies moléculaires pour lier, étaler spatialement et maintenir la manipulation. Les systèmes décrits ici comprennent, selon certains modes de réalisation, trois modules fonctionnels qui assurent la médiation d'opérations m6A : (1) un module initiateur synthétique pour placer m6A à des sites génomiques spécifiques; (2) un module de lecture synthétique pour programmer la reconnaissance m6A et la logique transcriptionnelle dépendant de m6A; et (3) un module de propagation qui met en oeuvre une lecture-écriture, un mécanisme proposé pour souligner l'étalement de la chromatine et le maintien épigénétique à travers les systèmes cellulaires.
