(JA) 多能性幹細胞の分化制御方法
Abstract:
(EN) This differentiation control method for pluripotent stem cells is characterized by: selecting laminin or a fragment thereof using binding affinity to the pluripotent stem cells as an index, the binding affinity being determinable by temporally observing the survival rate and mobility of the cells; and inducing the differentiation of the pluripotent stem cells in the presence of the laminin or a fragment thereof. The present invention allows easy production, from pluripotent stem cells, of a cell population in which the ratio of differentiation-induced cells is arbitrarily changed. A cell population obtained by the production method is highly useful for treatment of diseases by cellular therapy.
(FR) Cette invention concerne un procédé de contrôle de différenciation pour cellules souches pluripotentes caractérisé par : la sélection de la laminine ou d'un fragment de celle-ci à l'aide de son affinité de liaison aux cellules souches pluripotentes à titre d'indice, où l'affinité de liaison peut être déterminée par observation dans le temps du taux de survie et de la mobilité des cellules ; et l'induction de la différenciation des cellules souches pluripotentes en présence de laminine ou d'un fragment de celle-ci. La présente invention permet la production facile, à partir de cellules souches pluripotentes, d'une population cellulaire où le taux de cellules induites par différenciation peut être changé de manière arbitraire. La population cellulaire obtenue par le procédé de production selon l'invention est très utile pour le traitement des maladies par thérapie cellulaire.
(JA) 多能性幹細胞との結合親和性、ここで結合親和性は細胞生着率及び細胞の移動性を経時観察することで判断することができるが、これを指標としてラミニン又はそのフラグメントを選択し、該ラミニン又はそのフラグメントの存在下に多能性幹細胞を分化誘導することを特徴とする、多能性幹細胞の分化制御方法。本発明により、分化誘導細胞の比率を任意に変化させた細胞集団を多能性幹細胞から簡便に製造することができる。当該製造方法により得られる細胞集団は、細胞医療による疾患の治療に極めて有用である。
