Abstract:
(EN) Described herein are methods and kits to detect the methylation state of a CpG dinucleotide in methylene tetrahydrofolate reductase (MTHFR) and uses thereof. In some embodiments the methods can include the steps of providing a biological sample from the subject; contacting DNA from the biological sample with bisulfite under alkaline conditions to produce bisulfite-treated DNA; contacting the bisulfite-treated DNA with a first oligonucleotide probe, wherein the first oligonucleotide probe is complementary to a nucleotide sequence that includes a CpG dinucleotide in the methylene tetrahydrofolate reductase (MTHFR) gene, wherein the first oligonucleotide probe detects either the unmethylated CpG dinuclotide or the methylated CpG dinucleotide; and detecting either the unmethylated CpG dinucleotide or the methylated CpG dinucleotide in the MTHFR gene, wherein methylation of the CpG dinucleotide in the MTHFR gene is associated with amplification of response of the reporter CpG indicating the exposure of the subject to the MMA.
(FR) L'invention concerne des procédés et des kits permettant de détecter l'état de méthylation d'un dinucléotide CpG dans la méthylène tétrahydrofolate réductase (MTHFR) et leurs utilisations. Selon certains modes de réalisation, les procédés peuvent comprendre les étapes consistant à fournir un échantillon biologique provenant du sujet ; à mettre en contact l'ADN de l'échantillon biologique avec du bisulfite dans des conditions alcalines pour produire de l'ADN traité au bisulfite ; à mettre en contact l'ADN traité au bisulfite avec une première sonde oligonucléotidique, la première sonde oligonucléotidique étant complémentaire d'une séquence nucléotidique qui comprend un dinucléotide CpG dans le gène de la méthylène tétrahydrofolate réductase (MTHFR), la première sonde oligonucléotidique détectant soit le dinucléotide CpG non méthylé, soit le dinucléotide CpG méthylé ; et à détecter soit le dinucléotide CpG non méthylé, soit le dinucléotide CpG méthylé dans le gène de la MTHFR, la méthylation du dinucléotide CpG dans le gène de la MTHFR étant associée à l'amplification de la réponse du CpG rapporteur indiquant l'exposition du sujet au MMA.
