Abstract

Provided is a method of inserting a polynucleotide sequence into a genome of a cell. The method comprises: generating a double-strand break at a target site of the genome; and introducing into the cell a virus. The virus comprises a nucleic acid comprising the polynucleotide sequence to be inserted or the complementary sequence thereof. The nucleic acid does not comprise a homologous arm or comprises very short (5-25 bp) homologous arms corresponding to the target site. Also provided herein is a composition for inserting a polynucleotide sequence into a genome of a cell. The composition comprises a site-specific nuclease capable of generating a DNA double-strand break at a target site of the genome and a virus comprising a nucleic acid comprising the polynucleotide sequence or the complementary sequence thereof.

L'invention concerne un procédé d'insertion d'une séquence polynucléotidique dans un génome d'une cellule. Le procédé comprend : la génération d'une cassure double brin au niveau d'un site cible du génome; et l'introduction d'un virus dans la cellule. Le virus comprend un acide nucléique comprenant la séquence polynucléotidique à insérer ou la séquence complémentaire de celle-ci.. L'acide nucléique ne comprend pas de bras homologue ou comprend des bras homologues très courts (5-25 bp) correspondant au site cible. L'invention concerne également une composition pour insérer une séquence polynucléotidique dans un génome d'une cellule. La composition comprend une nucléase spécifique à un site capable de générer une cassure double brin d'ADN au niveau d'un site cible du génome et un virus comprenant un acide nucléique comprenant la séquence polynucléotidique ou la séquence complémentaire de celle-ci.
