Abstract:
(EN) Provided is a single-domain antibody capable of recognizing human serum albumin (HSA). By adopting a phage display technology, multiple strains of antibodies capable of specifically recognizing HSA are obtained by screening in a human single-domain antibody library. The antibodies contain only one variable region of an antibody heavy chain and are the smallest fully functional antigen-binding fragments, and have a smaller molecular weight and weaker immunogenicity than a single-chain antibody or antibody Fab fragment, and can function to extend the half-life of biological drugs when specifically bound to HSA. Single-domain antibodies with a high affinity are obtained by screening, and the antibodies can recognize specific human serum albumin and extend the half-life of biological drugs, facilitating the treatment of tumors.
(FR) L'invention concerne un anticorps à domaine unique capable de reconnaître l'albumine sérique humaine (HSA). Selon l'invention, de multiples souches d'anticorps capables de reconnaître de manière spécifique la HSA sont obtenues par criblage dans une bibliothèque d'anticorps à domaine unique humain, en adoptant une technologie d'exposition sur phage. Les anticorps contiennent uniquement une région variable d'une chaîne lourde d'anticorps et sont les plus petits fragments de liaison à l'antigène complètement fonctionnels, et ont un poids moléculaire plus petit et une immunogénicité plus faible qu'un anticorps à chaîne unique ou un fragment Fab d'anticorps, et peuvent fonctionner pour prolonger la demi-vie de médicaments biologiques lorsqu'ils sont spécifiquement liés à la HSA. Selon l'invention, des anticorps à domaine unique ayant une affinité élevée sont obtenus par criblage, et les anticorps peuvent reconnaître l'albumine sérique humaine spécifique et peuvent prolonger la demi-vie de médicaments biologiques, facilitant ainsi le traitement de tumeurs.
