Abstract:
(EN) Disclosed herein is a nanoparticle comprising a copolymer comprising a polyethylene glycol polymer, a polyhydroxylated polymer, and a peroxide-sensitive pendant group; a glucose-responsive agent; and a therapeutic agent; wherein the copolymer encapsulates the glucose-responsive agent and the therapeutic agent. Also disclosed herein is a method of delivering a therapeutic agent to a subject comprising administering to the subject a nanoparticle comprising a copolymer comprising a polyethylene glycol polymer, a polyhydroxylated polymer, and a peroxide-sensitive pendant group; a glucose-responsive agent; and a therapeutic agent; wherein the copolymer encapsulates the glucose-responsive agent and the therapeutic agent; and releasing the therapeutic agent from the nanoparticle in the presence of hyperglycemic levels of glucose. In some embodiments, the glucose-responsive agent produces a peroxide when exposed to hyperglycemic levels of glucose, thereby triggering disassembly of the nanoparticle and release of encapsulated therapeutic agent.
(FR) L'invention concerne une nanoparticule comprenant un copolymère comprenant un polymère de polyéthylène glycol, un polymère polyhydroxylé et un groupe pendant réactif au peroxyde; un agent sensible au glucose; et un agent thérapeutique, le copolymère encapsulant l'agent sensible au glucose et l'agent thérapeutique. L'invention concerne également un procédé de traitement d'un sujet par un agent thérapeutique, comprenant l'administration au sujet d'une nanoparticule comprenant un copolymère comprenant un polymère de polyéthylène glycol, un polymère polyhydroxylé et un groupe pendant réactif au peroxyde; un agent sensible au glucose; et un agent thérapeutique, le copolymère encapsulant l'agent sensible au glucose et l'agent thérapeutique; et la libération de l'agent thérapeutique de la nanoparticule en présence de taux hyperglycémiques de glucose. Dans certains modes de réalisation, l'agent sensible au glucose produit un peroxyde lorsqu'il est exposé à des taux hyperglycémiques de glucose, ce qui permet de déclencher le désassemblage de la nanoparticule et la libération de l'agent thérapeutique encapsulé.
