Abstract:
(EN) A system, methods, and apparatus are described to collect and prepare single cells, nuclei, subcellular components, and biomolecules from specimens including tissues. The system can perform enzymatic and/or physical disruption of the tissue to dissociate it into single-cells or nuclei in suspension or subcellular components including nucleic acids. In some embodiments, the titer of dissociated cells is monitored at intervals and the viability determined. In some embodiments, the processing is adjusted according to the measurements of the titer and viability. In some embodiments, the single-cells or nuclei in suspension are washed and resuspended in the buffer or media of choice. In some embodiments, the conditions are chosen to produce nuclei. In other embodiments, the single-cells or nuclei are purified by affinity paramagnetic bead processing. In some embodiments, matched bulk nucleic acid to the single-cells is produced. In other embodiments, single-cell libraries, or nuclei libraries, or matched bulk libraries, or bulk libraries are produced. The single cells or nuclei can then be further processed by FACS, DNA sequencing, mass spectrometry, fluorescence, or other methods. In other embodiments, the tissue processing is integrated with an analytical system to produce a sample-to-answer system such as a tissue-to-genomics system.
(FR) L'invention concerne un système, des procédés et un appareil pour collecter et préparer des cellules isolées, des noyaux, des constituants sous-cellulaires et des biomolécules à partir d'échantillons comprenant des tissus. Le système peut effectuer une destruction enzymatique et/ou physique du tissu pour le dissocier en cellules isolées ou en noyaux en suspension ou en constituants sous-cellulaires comprenant des acides nucléiques. Dans certains modes de réalisation, le titre en cellules dissociées est surveillé à des intervalles et la viabilité est déterminée. Dans certains modes de réalisation, le traitement est ajusté en fonction des mesures du titre et de la viabilité. Dans certains modes de réalisation, les cellules isolées ou les noyaux en suspension sont lavés et remis en suspension dans le tampon ou le milieu choisi. Dans certains modes de réalisation, les conditions sont choisies pour produire des noyaux. Dans d'autres modes de réalisation, les cellules isolées ou les noyaux sont purifiés par traitement d'affinité par billes paramagnétiques. Dans certains modes de réalisation, un acide nucléique global apparié aux cellules isolées est produit. Dans d'autres modes de réalisation, des bibliothèques de cellules isolées ou des bibliothèques de noyaux ou des bibliothèques globales appariées ou des bibliothèques globales sont produites. Les cellules isolées ou les noyaux peuvent ensuite en outre être traités par FACS (tri de cellules activées par fluorescence), séquençage d'ADN, spectrométrie de masse, fluorescence ou d'autres procédés. Dans d'autres modes de réalisation, le traitement de tissu est intégré dans un système analytique pour produire un système échantillon-à-réponse tel qu'un système de tissu-à-génomique.
