Abstract:
(EN) The present invention provides methods for isolating human cardiac ventricular progenitor cells (HVPs), wherein cultures of day 5-7 cardiac progenitor cells are negatively selected for one or more first markers expressed on human pluripotent stem cells, such as TRA- 1-60, to thereby isolate HVPs. The methods can further include positive selection for expression of a second marker selected from the group consisting of JAG1, FZD4, LIFR, FGFR3 and TNFSF9. Large populations, including clonal populations, of isolated HVPs that are first marker negative/second marker positive are also provided. Methods of in vivo use of the HVPs for cardiac repair or to improve cardiac function are also provided. Methods of using the HVPs for cardiac toxicity screening of test compounds are also provided.
(FR) La présente invention concerne des procédés permettant d'isoler des cellules progénitrices ventriculaires cardiaques humaines (HVP), des cultures de cellules progénitrices cardiaques ayant 5 à 7 jours étant soumises à une sélection négative pour un ou plusieurs premiers marqueurs exprimés sur des cellules souches pluripotentes humaines, telles que TRA-1-60, pour isoler ainsi les HVP. Les procédés peuvent comprendre en outre une sélection positive pour l'expression d'un second marqueur choisi dans le groupe constitué de JAG1, FZD4, LIFR, FGFR3 et TNFSF9. L'invention concerne également de grandes populations, y compris des populations clonales, de HVP isolées qui sont négatives au premier marqueur/positives au second marqueur. L'invention concerne également des procédés d'utilisation in vivo des HVP à des fins de réparation cardiaque ou pour améliorer la fonction cardiaque. L'invention concerne également des procédés d'utilisation des HVP à des fins de dépistage de la toxicité cardiaque de composés testé.
