Abstract:
(EN) The present invention relates to a method for obtaining neural stem cells and neural progenitor cells (NSC/NPC) from ovarian cortical tissue of pre-pubescent female mammals, without needing to add factors for inducing and/or expanding the neural line to the culture medium. The method of the invention can be used to obtain NSC/NPC after 8–21 days of incubation, improving the results by adding follicle stimulating hormone (FSH) to the culture medium. The NSC/NPC obtained using this method can be used in experimental models of in-vitro neurogenesis, in directed NSC/NPC differentiation to generate different types of nerve cells of interest in medicine for regenerating the nervous system, with use in autologous transplantation in cell therapy of neurodegenerative diseases, and to evaluate the neurotoxicity of different drugs, contaminants and chemical compounds.
(FR) La présente invention se rapport à un procédé d’obtention de cellules souches neurales et de cellules progénitrices neurales (CSN/CPN) à partir de tissu cortical ovarien de femelles prépubères de mammifères, sans qu’il soit nécessaire d’ajouter des facteurs d’induction et/ou d’expansion de la lignée neurale au milieu de culture. Le procédé de l’invention permet d’obtenir les CSN/CPN après 8-21 jours d’incubation, ce qui améliore les résultats par rapport à ceux obtenus avec l'addition de FSH au milieu de culture. Les CSB/CPN obtenues par ce procédé peuvent être utilisées comme modèles expérimentaux de la neurogenèse in vitro, pour une différenciation dirigée des CTN/CPN afin produire divers types de cellules nerveuses d’intérêt en médecine régénérative du système nerveux, trouvant application dans des greffes autologues au cours de thérapies cellulaires de pathologies neurodégénératives, ainsi que pour l’évaluation de la neurotoxicité de divers produits pharmaceutiques, contaminants et composés chimiques.
(ES) La presente invención se refiere a un método para obtener células troncales neurales y células progenltoras neurales (CTN/CPN) a partir de tejido cortical ovárico de hembras prepúberes de mamífero, sin necesidad de añadir al medio de cultivo factores de inducción y/o expansión del linaje neural. Con el método de la invención se obtienen CTN/CPN tras 8-21 días de incubación, mejorando los resultados mediante la adición de FSH al medio de cultivo. Las CTN/CPN que se obtienen mediante este método pueden ser utilizadas en modelos experimentales de neurogénesis in vitro, en diferenciación dirigida de CTN/CPN para generar diversos tipos de células nerviosas de interés en medicina regenerativa del sistema nervioso, con aplicación al trasplante autólogo en terapia celular de patologías neurodegenerativas, así como para la evaluación de neurotoxicidad de diversos fármacos, contaminantes y compuestos químicos.
